چکیده

Although type E strains of Clostridium botulinum have been implicated in human botulism since 1936, no studies of the isolation or characterization of the toxin have been reported. Crude toxoids have been prepared by various workers for immunization of animals and preparation of antitoxin. Barron and Reed (1954) presented a systematic study of methods for preparation of crude alum precipitated type E toxoid, employing cultures grown in cellophane dialysis bags. Publications from our laboratory have described procedures for production and purification of C. botulinum types A, B, and C toxins suitable for preparations of purified toxoids (Duff et al., 1957a, 1957b; Cardella et al., 1956). Duff et al. (1956) reported that the intraperitoneal toxicity for mice of crude and partially purified type E toxin was increased strikingly when the culture was incubated with trypsin. The present paper describes an extension of these studies to the preparation of partially purified type E toxoid. The purification and detoxification of nonactivated and trypsin activated type E toxins were compared, and the immunization of mice by aluminum phosphate toxoids was investigated.2

Double Sandwich ELISA Modified Method for the Detection of Clostridium Botulinum Type E

Background & Objective: A very small amount of botulinum toxin can cause death and on the other hand, there is no cure for its poison other than antitoxin. Therefore, a diagnostic method that can detect very small amounts of botulinum toxin in a short time is very important. In this study, rapid and accurate detection of botulinum toxin type E has been performed with the double sandwich ELISA m...

Purification and some properties of progenitor toxins of Clostridium botulinum type B.

Purification of progenitor toxin of Clostridium botulinum type B strain Okra was undertaken by sequential steps of acid precipitation, extraction, ammonium sulfate precipitation, ribonuclease digestion, acid precipitation, protamine treatment, sulphopropyl-Sephadex chromatography, and Sephadex G-200 gel filtration. Two different molecular-sized toxins, named large (L) and medium (M) toxins, wer...

Characterization of Clostridium botulinum spores and its toxin in honey

Botulism is a serious paralytic disease caused by Clostridium botulinum toxin in foods. There are seven recognized serotypes of botulinum neurotoxins among which the principal prevalent types in humans include A, B and E. Infant botulism results from intestinal colonization and toxin production by C. botulinum spores in babies less than 1 year old. Honey is the most important food discriminated...

Comparison of toxins of Clostridium butyricum and Clostridium botulinum type E.

The toxin of Clostridium butyricum strains isolated from two infants with botulism is neutralized by antitoxin for type E botulinum toxin. This toxin and that of a C. botulinum type E strain were purified by the same protocol. Both toxins were Mr 145,000 proteins which, when activated with trypsin, were composed of an H subunit of Mr 105,000 and an L subunit of Mr 50,000. The activated specific...
